1. Ready-to-go Lentiviral Preparation (quantitatively titrated).

    We will provide ready to be used lentiviral preparations of the following types:

            a. hPGK - GFP concentrated lentiviral soup (paints your cells in green).

                This lentivirus expresses eGFP under the strong constitutive hPGK promoter.

            b. hPGK - mCherry concentrated lentiviral soup (paints your cells in red).

                This lentivirus expresses mCherry under the strong constitutive hPGK promoter.

    New additions (due to popular demand)

            c. pLKO.1-scrambled shRNA concentrated lentiviral soup.*, **

                This lentivirus serves as negative control for lenti made with Sigma-Aldrich pLKO.1 shRNA plasmids.

            d. pGIPZ-scrambled shRNA concentrated lentiviral soup.*

                This lentivirus serves as negative control for lenti made with Open Biosystems pGIPZ shRNA plasmids.

    *BIMR Viral Vectors has purchased the above plasmids from Sigma-Aldrich and Open Biosystems and does not profit from

     the sale of lentivirus in any way. All lentiviral vectors produced by BIMR Viral Vectors and distributed to BIMR laboratories

     are for research use only. The Burnham Institute for Medical Research is in a partnership program with both Sigma-Aldrich

     and Open Biosystems. Researchers are encouraged to purchase their shRNA lenti constructs from either company.

    **pLKO.1 vectors do not have a fluorescent marker. These ready-to-go lenti preparations are not titrated, however, their titer

    is expected to be equally high as fluorescent lentiviral soups (~10⁸ i.u./mL).

    Your 300μL lentiviral prep will be aliqoted in 0.65mL microcentrifuge tubes (standard = 30μL/tube) and stored @ -80°C.

2. Custom Packaged Lentiviral Preparation (especially useful with shRNA - see useful literature).

    We will produce lentiviral particles using the custom second / third generation transfer vector which you provide (see guidelines).

    All packaging plasmids (pCMVΔR8.74; pMD.G (VSVG)) are provided by BIMR Lentiviral Core.

    Your 300μL lentiviral prep will be aliqoted in a 0.65mL microcentrifuge tubes (30μL/tube) and stored @ -80°C.

Your Options for Lentiviral Preparations (1. and 2.) are as follows:

1. Unfiltered, Unconcentrated Virus Production as VSVG Pseudotyped Viral Supernatants.

2. Filtered, Concentrated Virus Production as VSVG Pseudotyped Viral Supernatants.

    Concentration is performed by Ultracentrifugation.

3. Quantitative Determination (using fluorescent microscopy) of viral titers using 293T cells.*

    (N.B.: all but one Ready-to-Go Preps are quantitatively titrated).

            *BIMR Viral Vectors performs quantitative titrations only on 293T cells if the viral vector contains a fluorescent marker.

                Please do NOT request titrations on other cell lines or using methods other than fluorescence microscopy.

3. Mycoplasma Testing

    We will test any of your cell cultures for mycoplasma (see guidelines) using PCR on a highly conserved region of the mycoplasma DNA.

    N.B.: All of our 293T cells producing your virus are routinely tested for mycoplasma to ensure that you will receive non-contaminated preps.
